A equipe santista chega ao confronto com vantagem após vencer o primeiro jogo por 2 a 0, na Vila Belmiro. Com o resultado, o Santos pode até perder por um gol de diferença para avançar à próxima fase.

A classificação representaria uma mudança no retrospecto do clube na competição. Em nove participações anteriores, o Santos nunca conseguiu ultrapassar as quartas de final. A primeira vez que o Peixe alcançou essa fase foi em 2003, justamente em sua estreia na Sul-Americana. O adversário era o Cienciano, do Peru. No jogo de ida, na Vila Belmiro, as equipes empataram por 1 a 1. Na partida de volta, disputada em Cusco, os peruanos venceram por 2 a 1 e eliminaram o Santos.
O Cienciano, inclusive, ainda pode aparecer no caminho do Peixe nesta edição. A equipe peruana enfrenta o Montevideo City Torque, do Uruguai, pelas quartas de final. No primeiro confronto, realizado em Cusco, o Cienciano venceu por 2 a 0.
Em 2004, o Santos voltou a alcançar as quartas de final. Dessa vez, o adversário foi a LDU, do Equador. O clube brasileiro perdeu os dois jogos: 3 a 2 fora de casa e 2 a 1 na Vila Belmiro.
A terceira participação do Santos nas quartas aconteceu em 2021, contra o Libertad, do Paraguai. Na Vila Belmiro, o Santos venceu o Libertad por 2 a 1. Carlos Sánchez abriu o placar para o time brasileiro, enquanto Bocanegra deixou tudo igual para os paraguaios.
Nos acréscimos do segundo tempo, o zagueiro Alexander Barboza, atualmente no Palmeiras, marcou contra e garantiu a vitória santista. A vantagem, porém, não foi suficiente para o Peixe avançar. No jogo de volta, o Libertad venceu por 1 a 0, com gol de Sebastián Ferreira, atacante que passou pelo Vasco em 2023.
O placar agregado terminou empatado em 2 a 2. Naquele ano, o regulamento ainda previa o gol marcado fora de casa como critério de desempate, e o Libertad ficou com a vaga.
Décima participação pode marcar mudança no retrospecto
Depois de nove campanhas, o Santos chega à sua décima participação na Copa Sul-Americana. O clube disputou o torneio também em 2005, 2006, 2010, 2019, 2022 e 2023.
Em 2005, a eliminação aconteceu na primeira fase, contra o Fluminense. No ano seguinte, o Peixe chegou às oitavas de final, mas foi eliminado pelo San Lorenzo, da Argentina. Em 2010, o Santos caiu na segunda fase diante do Avaí. Já em 2019, a eliminação ocorreu na primeira fase, contra o River Plate, do Uruguai.
Após voltar às quartas em 2021, o clube foi eliminado nas oitavas de final em 2022 pelo Deportivo Táchira, da Venezuela. Na última participação, em 2023, o Santos não avançou da fase de grupos e terminou na terceira colocação, atrás de Newell’s Old Boys, da Argentina, e Audax Italiano, do Chile.
